Abstract
The characterization and quantification of proteins in MALDI spectra were improved using differential isotope labeling. Peptide mass fingerprints were acquired on a Voyager DE-STR reflector MALDI-ToF mass spectrometer. All product ion spectra were acquired on a Q-ToF I instrument equipped with a dual electrospray source. The collision energy was optimized for each peptide analyzed. The results show that guanidination chemistry allows relative quantification to be carried out with additional benefits in protein identification through improved MALDI MS spectra and easing interpretation of tandem mass spectra.
