Abstract

Expressed protein ligation (EPL) is a protein semi-synthesis approach that allows synthetic and recombinant polypeptides to be chemoselectively and regioselectively joined together via a peptide bond. The approach makes the primary structure of proteins accessible to the tools of synthetic organic chemistry and thus provides expanded opportunities in the area of protein engineering. The development and application of this process will be discussed.
