A modular cross-linking approach for exploring protein interactions

Abstract

The protein-protein interaction was described using cross-linking reagents and mass spectrometry. This approach was applied for the study of cross-linking between the components of the heterodimeric protein complex, Negative Cofactor 2 (NC2). Five crosslinking reagents were synthesized using solid phase chemistry, by availing the chemical versatility of the mercaptopropionamide-PEGA resin-linker. It was found that there is an intermolecular interaction between the N-terminal regions of NC2 α and β, as well as three intra-molecular and several dead-end cross-links.
